COSMIC v100: Tracing our origins with a re-curation of BRAF and RAS genes

It has been 20 years since COSMIC was first launched, yet our journey to fully understanding cancer genes is far from over. Researchers are still making discoveries on the role of BRAF and RAS genes in cancer and figuring out how to treat tumors driven by mutations in these genes. 

With this in mind, we trace our origins with another deep curation on the first four COSMIC genes – BRAF, KRAS, HRAS and NRAS – among other improvements.

Substantial update to BRAF & RAS genes
  • 1356 new whole genomes 
  • 29 new systematic screen papers 
  • 241 new publications 
  • Over 307k new genomic variants 
New additions
  • Drug resistance pairs: EGFR–Cetuximab, IDH2–Enasidenib and KRAS–Agarasib 
  • Cancer Gene Census (CGC) genes: RXRA (Tier 1) and NR2F2, PIK3R3, PLEC and TNRC18 (Tier 2) 
  • Hallmarks of Cancer annotations for 8 CGC Tier 1 genes: TRRAP, TSC1, TSC2, U2AF1, WT1, MYCN, ENT5C and ARGHAP35 
Adjustments
  • CBLC reassigned to Tier 2 of CGC 
  • Cancer Mutation Census (CMC) updated to align with the latest COSMIC release 

This release also marks the official phasing out of the old download files.
